# Glioblastoma

Glioblastoma is a plausible but still speculative setting for WFA.

The biology makes sense. The translational evidence remains limited.

### Why glioblastoma is of interest

GBM depends heavily on survival networks such as `HSP90`, `Akt/mTOR`, stem-like persistence, and treatment resistance. Those are all areas where WFA has mechanistic relevance.

### Cell data

In glioblastoma models, WFA has produced G2/M arrest and apoptosis, with effects linked to `Akt/mTOR` suppression and proteostasis disruption.

It has also shown radiosensitising potential and some anti-stem-cell relevance in patient-derived GBM stem-like systems.

### Animal data

Animal evidence exists but is thinner than in breast, ovarian, or lung cancer.

That means glioblastoma remains a biologically credible area rather than a clinically prioritised one.

### Blood-brain barrier considerations

This is the main translational challenge.

Standard WFA formulations are unlikely to achieve reliable therapeutic CNS exposure through an intact blood-brain barrier. GBM does involve abnormal and leaky tumour vasculature, which may help somewhat, but this remains uncertain.

Preclinical nanoformulations and local-delivery concepts have been explored, but they are not clinically established.

### Clinical setting assessment

The unmet need in GBM is high, and the mechanism fit is real. That still does not overcome the limited in vivo depth and unresolved CNS delivery problem.

### Bottom line

Glioblastoma is worth following mechanistically, but it should not be over-prioritised clinically at this stage.
