# HER2CLIMB Guide

### What HER2CLIMB is

HER2CLIMB is the name of the pivotal tucatinib trial and the regimen that grew out of it: **tucatinib + trastuzumab + capecitabine** for previously treated **HER2-positive metastatic breast cancer**.

It emerged in the late 2010s as a key study in patients who had already received earlier HER2-directed therapy.

### What the main findings showed

So far, HER2CLIMB is best known for showing that adding **tucatinib** to trastuzumab and capecitabine improved outcomes compared with trastuzumab and capecitabine alone.

The most important findings were:

* improved **progression-free survival**
* improved **overall survival**
* meaningful benefit in patients with **brain metastases**

That is why the regimen became an important later-line option in HER2-positive metastatic breast cancer, especially when CNS disease is part of the picture.
